ABSTRACT

Objective:To analyze the horizontal scientific research projects from 2015 to 2019 by the provincial Centers for Disease Control and Prevention (CDC) in China, and to compare the regional differences, in order to provide the suggestion on the scientific management of CDC.Methods:The horizontal scientific research projects from 2015 to 2019 were retrospectively analyzed by questionnaire survey. Multiple linear regression models were adopted to examine the trend, and variance analyses were used to test the differences in horizontal scientific research projects among the Eastern, Central, and Western regions.Results:From 2015 to 2019, provincial CDC have received RMB 124.3 million of horizontal scientific research project funds totally, of which 51.9% were funded by enterprises, and 86.9% were undertaken by provincial CDC themselves. There were no statistical significance in the change of research project funds obtained by provincial CDC ( F=0.46, P = 0.764) during this period.The number of horizontal scientific research projects undertook or participated by provincial CDC in the Eastern region were more than that of the Central and Western region ( F = 5.85, P = 0.004; F = 5.03, P = 0.008). Conclusions:The horizontal scientific research projects obtained by the provincial CDC remained stable in recent years while distribution was unbalanced in the region areas. It is suggested to innovate the management mode of scientific research projects with strengthening the trans-agency, trans-department and trans-regional cooperation.

ABSTRACT

Objective:To analyze the drug resistance and molecular epidemiological characteristics of Acinetobacter baumannii isolated from pediatric patients. Methods:Acinetobacter baumannii isolated from patients hospitalized in Inner Mongolia Medical University Affiliated Hospital from January 2016 to June 2018 was collected.Vitek-2 Compact automatic microbiological identification and drug sensitivity analysis system was used to identify and test the drug sensitivity of Acinetobacter baumannii isolates, and pulse field gel electrophoresis (PFGE) and multilocus sequence analysis (MLST) were applied to the homology analysis of the strains. Results:A total of 94 clinical isolates of Acinetobacter baumannii were collected, of which 42 strains were isolated from pediatric patients and 52 strains from adult patients.The drug resistance rates of pediatric isolates to Imipenem, and Meropenem and Tigecycline were 7.1%, 7.1% and 0, respectively, and the drug resistance rates of adult isolates to these 3 antibiotics were 67.3%, 54.8%, and 5.5%, respectively.The results of PFGE typing showed that 94 strains were divided into 49 genotypes (X1-X49 type), 52 adult strains were distributed in 22 genotypes, and 42 pediatric strains were distributed in 33 genotypes.The dominant genotype was X23 (21 strains, 22.3%), of which 18 strains(85.7%) were adult isolates and 3 strains (14.3%) were children isolates.The drug resistance rate of X23 genotypes to carbapenems was 100%, which was significantly higher than that of other genotypes.The results of MLST genotyping showed that X23 genotype was ST195, which belonged to clonal complex(CC92) clone. Conclusions:The overall drug resistance rate of Acinetobacter baumannii isolates in Inner Mongolia Medical University Affiliated Hospital was significantly lower than that of adult isolates, and the diversity of genotypes was obvious.The dominant genotypes of the strains belongs to the CC92 clone population, and is the dominant clone strain in many places of our country.

ABSTRACT

Objective This paper aimes to analyze the scientific research development trend,research emphasis and cooperation situation of Chinese Center for Disease Control and Prevention since its establishment,to provide the suggestions for policy making of scientific management and related decisions.Methods Based on GoPubMed,Literature quantitative analysis was used to analyze the publication time and journal distribution,research topics and core authors of the scientific papers from 2002 to 2015.Results A total number of 4 501 research papers had been published in 576 different kinds of periodicals.Research topics were based on population health with special focus on the epidemiological,microbiological and etiological studies of infectious disease.Chronic non-communicable disease and health research were paying more and more attention to.Core author group and the core research team have been formed.Conclusions The capability to conduct scientific research was increased and the international influence was increasing year by year,the ability of infectious disease control was strengthened,and the scientific research team worked together more closely.Thus,the Chinese Center for Disease Control and Prevention should reinforce scientific management,develop advanced disciplines,as well as improve the quality of scientific research in the future.

ABSTRACT

Objective:To study influence study of near and long term survival rate on different resection regions for patients with gastric cancer in M region.Methods:110 patients with M region gastric cancer received treatment in our hospital were divided into observation group(with total gastrectomy) 57 cases and control group (with subtotal gastrectomy) 53 cases,compared with clinical pathology data and radical degree,two groups of patients' operation index,and local recurrence rate and near and long term survival rate in the two groups.Results:Two groups of patients were compared with clinical pathology data and radical degree,the difference was not statistically significant (P>0.05).The observation group's nearly cutting edge outside distance and lymph nodes were significantly higher than the control group,the difference was statistically significant (P<0.05).There was no significant difference between the two groups(P>0.05).The recurrence rate(3.51%) and recurrence rate (15.79%) of the observation group were significantly less than the control group of 16.98% and 33.96%,and the difference was statistically significant (P<0.05).The 1 and 3 year survival rates of the observation group were compared with the control group,the difference was not statistically significant (P>0.05).But the 5 year survival rate of the observation group was 57.89%,which was significantly higher than that in the control group of 35.85%,the difference was statistically significant (P<0.05).Conclusions There is no significant difference in the survival rate of total gastrectomy in patients with gastric cancer in M region and total gastrectomy,but the long-term survival rate is higher,it is worthy of clinical attention.

ABSTRACT

Objective To investigate the status of work-family balance,and explore the relationship between work-family balance and well-being of female prison police.Methods 185 female prison police from one of prison of Sichuan province were assessed using the questionnaires of work-family balance ques tionnaire and well-being scale,and the data were analyzed using t test,one-way ANOVA,pearson correlation analysis and logical multiple linear regression analysis.Results The total score of work-family balance was (10.22± 1.32).The dimension score of work-family balance from high to low were:resource sharing(3.44± 0.60),family balance (3.38±0.53) and work balance (3.12±0.60).There was no remarkable difference on work-family balance between stage of age and family monthly income;but work-family balance declined with the increases of marriageable age and working age (F=13.01,8.25,both P< 0.01).Work balance,family balance resource sharing,work-family balance had significant positive correlation with well-being and it's dimensions(r=0.15 ~ 0.60,P<0.05).Regression analysis showed that age,marriageable age had a negative predic tive effect on job satisfaction and emotion satisfaction(β=-0.20,-0.24,both P<0.01),and work family balance and it's dimensions had a positive predictive effect on well-being(3=0.282~0.679,P<0.01).Conclusion Work-family balance has a significant predictive effect on well-being of female prison police.

ABSTRACT

Objective To explore the levels of neuron-speciifc enolase (NSE) of the cerebrospinal lfuid (CSF) in children with convulsion. Methods Ninety children with convulsion were enrolled. According to the frequency of convulsion attack, the children were divided into brief convulsion group 51 cases and prolonged convulsion group 39 cases, further, based on the etiology, the children were divided into viral encephalitis (VE) group, idiopathic epilepsy (EP) group, and febrile convulsion (FS) group. CSF was collected within 24-48 h convulsion attack. Twenty-three children with elective surgery were selected as a control group. CSF was collected before surgery. The NSE level of CSF were measured by ELISA method and compared among groups. Results The NSE levels of CSF in prolonged convulsion group and brief convulsion group were signiifcantly higher than that in control group, while the NES levels of CSF in prolonged convulsion group were signiifcantly higher than that in brief convulsion group (all P0.05). Conclusions Convulsion contributed to higher NSE levers of CSF, especially in children with prolonged convulsion attack or with VE. The NSE level of CSF can be regarded as an early objective indicator of brain damage after convulsions.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate whether selectively stimulating β1-adrenergic receptor could inhibit high mobility group box 1 protein and attenuate myocardial ischemia/reperfusion(I/R) injury in rats.</p><p><b>METHODS</b>Eighty healthy male Sprague-Dawley (SD) rats were randomly divided into seven groups: (1) Sham operated group (SO); (2) Ischemia/reperfusion (I/R) group; (3) Dobutamine1 (5 µg×kg⁻¹ · min⁻¹) + I/R group; (4) Dobutamine2 (10 µg·kg⁻¹ × min⁻¹) + I/R group; (5) LY294002 (0.3 mg/kg) + Dobutamine2 + I/R group; (6) SB203580 (1 mg/kg) + Dobutamine2 + I/R group; (7) ZnPPIX (10 mg/kg) + Dobutamine2+I/R group. Rats were pretreated by saline, dobutamine, LY294002, SB203580 and ZnPPIX, respectively, then underwent myocardial I/R. Myocardial I/R injury and oxidative stress were assessed, and myocardial HO-1, NF-κB and HMGB1 expressions were measured by Western blot analysis.</p><p><b>RESULTS</b>Dobutamine significantly reduced the myocardial infarct size (P < 0.05), myocardial enzymes (LDH and CK) (P < 0.05) and proinfiammation cytokines (TNF-α and IL-6), reduced oxidative stress (MDA and SOD) in a dose-dependent manner (all P < 0.05). Meanwhile, dobutamine significantly and dose-dependently mediated the induction of HO-1 (P < 0.05), the expression of NF-κB (P < 0.05) and HMGB1 (P < 0.05). However, all the effects could be significantly reversed by co-treatment with LY294002, SB203580 and ZnPPIX (all P < 0.05).</p><p><b>CONCLUSIONS</b>Current study demonstrates that selectively stimulating β1-adrenergic receptor by dobutasmine could reduce rat myocardial I/R injury in vivo through promoting the induction of HO-1 and inhibiting HMGB1 release.</p>

ABSTRACT

Objective:To evaluate the clinical research papers published in recent years about tuberculosis , to understand the ethical review in biomedical journals in our country .Method:We analyzed clinical papers published in two journals that are famous in the field of tuberculosis since 2010 .Results:There were 33 articles included in our study .The number of informed consent is higher than that of ethical review .The descriptions of informed con-sents were not standardized in 12%articles.9%of articles did not mention the name of the ethics committee .Con-clusion:The attention degree to ethics of some editors and researchers should be improved .Pay more attention to medical ethical education , improve cognitive level of ethical issues , ensure the interests of the participants and im-prove the quality of research .

ABSTRACT

<p><b>OBJECTIVE</b>To explore the effects of Kindlin-2 RNA interference on vascular smooth muscle cells (VSMCs) migration, adhesion and β1-integrin as well as the relationship between Kindlin-2 and β1-integrin.</p><p><b>METHODS</b>Primary VSMCs were cultured, infected with Kindlin-2 siRNA lentiviral vectors.VSMCs were divided into three groups:the blank control group, the negative control group and the Kindlin-2 siRNA group. The ability of VSMCs migration was measured by Transwell experiment and wound healing assay. The ability of VSMCs adhesion to extracelluar matrix was determined by cell-extracelluar matrix adhesion assay. The Kindlin-2 and β1-integrin mRNA and protein expression levels were detected by real-time quantitative PCR and Western blot. Total β1-integrin and active β1-integrin expression on the surface of VSMCs was evaluated by flow cytometry.</p><p><b>RESULTS</b>The efficiency of Kindlin-2 siRNA lentivirus infected VSMCs was more than 90%. The number of VSMCs migration in the Kindlin-2 siRNA group was significantly lower than that of the blank control group and the negative group (all P < 0.05). Moreover, the distance of VSMCs migration was shorter than that of the blank control group and the negative group (all P < 0.05). The number of VSMCs adhesion to collagen I was less than that of the blank control group and the negative group (all P < 0.05). A(590 nm) of the Kindlin-2 siRNA group was also lower than that of the blank control group and the negative group (all P < 0.05). Compared with the blank control group, the expression level of Kindlin-2 mRNA in the Kindlin-2 siRNA group decreased 47% (P < 0.05), but the expression level of β1-integrin mRNA remained unchanged. The Kindlin-2 protein level in the Kindlin-2 siRNA group was lower than that of the blank control group and the negative group (all P < 0.05). β1-integrin protein level was similar among the three groups. Activated β1-integrin on the surface of VSMCs in the Kindlin-2 siRNA group was lower than that of the blank control group and the negative group (all P < 0.05).However, the expression level of total β1-integrin on the VSMCs surface was similar among the three groups.</p><p><b>CONCLUSION</b>Kindin-2 can regulate VSMCs migration and adhesion and activate β1-integrin on the surface of VSMCs.</p>

ABSTRACT

From 2006 to 2010,381 national or provincial projects were granted to Chinese Center for Disease Control and Prevention (Chinese CDC).According to the development plan of key dsciplines made by Chinese CDC, this study stratified the granted projects and analyzed their distribution in the four units (including 37 subunits),namely,infection disease,health,chronic disease and general technology platform. The results indicated that most of the important topics proposed by our center received financial support from the government,and the supported projects were mostly in the field of infectious disease and health.The results also revealed that the key projects proposed by Chinese CDC meet the practical requirements of disease control,and those projects covered the key scientific topics and key technologies.Meanwhile,the result reflected that Chinese CDC is very competitive as far as scientific research is concerned,but the research structure is not balanced among the 4 units.

ABSTRACT

Objective To observe the effect of nitric oxide (NO) on exflagellation of malaria parasite. Methods The level of parasitemia and gametocytemia in DBA/2 mice infected with Plasmodium yoelii 17XL was measured by scanning Giemsa-stained blood smears, and the NO level in culture supernatant of splenocytes was checked using Griess reaction. The mice were injected with different doses of NO donor (NOC5) on day 4 post-infection, and control mice were injected with NOC5 precursor. On day 6 post-infection, mice were injected with NOS inhibitor (L-NMMA), and control mice were injected with D-NMMA and PBS, respectively. Blood samples were collected from tail vein of mice before injection, 30 and 60 min after being injected with NOC5 and NOC5 precursor, 4 and 8 h after being injected with L-NMMA, D-NMMA, and PBS respectively. Exflagellation number of gametocytes in blood culture was counted under microscope. Results The NO level in culture supernatant of splenocytes from mice on day 4 and 6 post-infection was 16.5 mmol/L and 30.4 mmol/L, and exflagellation number was 11.33 and 0.66, respectively. The number of exflagellation in parasitized erythrocytes, obtained from mice on day 4 post-infection, was 5.33 and 2.66, respectively, 30 and 60 min after injection of 1 mg NO donor (NOC5), significantly lower than that of the control (P
